How glycine is thought to work
Glycine does two useful things around bedtime. It acts directly as a calming signal at glycine and NMDA receptors, and it appears to lower core body temperature slightly, which is part of the normal wind-down into sleep.
A 2015 study by Kawai and colleagues traced glycine’s sleep effect to NMDA receptors in the suprachiasmatic nucleus, the brain’s master clock, along with a small drop in core temperature and increased blood flow to the skin. That mechanism lines up with what people report: falling asleep a bit faster and waking feeling less foggy, rather than a heavy sedative knockout.

A companion line of work summarized by Bannai and Kawai (2012) reported better sleep satisfaction and less next-day sleepiness at the same dose. These are small, short studies measuring how people feel rather than large trials with objective sleep-lab endpoints, so read the tier as promising, not established.
The practical takeaway: start at 3 g in the hour before bed. There is little reason to jump higher for sleep, since the benefit in these trials showed up at 3 g and larger doses were not tested for better sleep specifically.
What to look for in a glycine supplement for sleep
