Magnesium glycinate is magnesium bound to the amino acid glycine, a chelated form known for being gentle on the stomach and well absorbed. For most healthy adults, you can take it with melatonin. There is no well-documented direct interaction between the two, and they’re frequently combined in commercial sleep blends. The main cautions are additive drowsiness and each ingredient’s separate medication interactions, which is why the answer is “usually fine, but check with your prescriber if you take other drugs.”

Why people stack the two
Magnesium and melatonin work on sleep through different mechanisms, which is part of why combining them appeals to people.
Melatonin is a hormone your pineal gland releases as darkness falls. It signals your body clock that it’s time to wind down. Taken as a supplement, it mainly helps with sleep timing: shortening how long it takes to drift off and shifting a delayed body clock earlier.
Magnesium plays a quieter, more structural role. It’s a cofactor for hundreds of enzymatic reactions, including ones that regulate the calming neurotransmitter GABA and the NMDA receptor. Magnesium is also involved in the enzymatic pathway your body uses to make its own melatonin. The theory behind stacking them is complementary: melatonin sets the timing, magnesium supports the underlying calm. The glycine in magnesium glycinate has its own mild pro-sleep signal in some early research, though that evidence is still thin.
What the evidence actually shows
Melatonin has moderate, fairly consistent evidence for sleep-onset problems. Magnesium’s sleep evidence is more limited and lower quality. Studies of the specific combination are scarce.

The trial closest to your question, Rondanelli and colleagues (2011), combined melatonin with magnesium and zinc in older adults with insomnia and saw improved sleep quality against placebo. It’s a small, three-ingredient study, so it can’t tell you magnesium glycinate plus melatonin alone will help you. But it does show the pairing was used safely in a controlled setting.
For magnesium on its own, a 2021 systematic review by Mah and Pitre rated the evidence as low certainty: a hint of benefit, not a settled case. Melatonin fares better: a 2013 meta-analysis by Ferracioli-Oda of 19 trials found it reliably shortened sleep onset, and a 2017 review by Auld and colleagues reached similar conclusions for adult sleep disorders. Treat magnesium glycinate as emerging support for sleep and melatonin as moderate.
Typical use and timing
Common practice, not a prescription:
Magnesium glycinate doses typically run 200–400 mg of elemental magnesium, taken 30–60 minutes before bed. The NIH Office of Dietary Supplements sets the tolerable upper limit for supplemental magnesium at 350 mg/day, above which loose stools and diarrhea become more likely.
For melatonin, lower is usually better. Many people do well on 0.5–3 mg, taken 30–90 minutes before their target sleep time. Higher doses raise the odds of next-day grogginess and vivid dreams.
Taking both in the same pre-bed window is fine for most people. A sensible approach is to start each ingredient at the low end separately, learn how your body responds, then combine.
Cautions and interactions
The pairing is low-risk for healthy adults, but each ingredient has separate interactions worth knowing.
Additive sedation. Both push toward sleep. Stacked, they may leave you groggier the next morning, particularly at higher melatonin doses. Never combine either with alcohol or other sedatives without medical guidance, and don’t drive until you know how the combination affects you.
Melatonin’s interactions. Melatonin can interact with blood thinners (warfarin), blood-pressure medication, diabetes drugs (it may affect glucose), immunosuppressants, and some anticonvulsants. It’s also not first-line for pregnancy or breastfeeding.
Magnesium’s interactions. Supplemental magnesium can bind certain antibiotics (tetracyclines, quinolones) and bisphosphonates, reducing their absorption. Separate doses by a few hours. People with reduced kidney function can accumulate magnesium to dangerous levels and should not supplement without supervision.
